Método de inoculação de Hemileia vastarix Berk. et Br. em folhas destacadas de cafeeiro

Abstract

Os estudos genéticos do cafeeiro e de Hemileia vastatrix são muitas vezes dificultados pela característica do fungo de multiplicar apenas nos tecidos da planta hospedeira. Assim, esse trabalho foi proposto com o objetivo de desenvolver um método de inoculação de H. vastatrix que permita estudar com segurança raças e a herança da resistência do cafeeiro à ferrugem. Para isso, utilizou-se a raça II de H. vastatrix, C. arabica ‘Catuaí’ como hospedeiro susceptível e a população segregante de cafeeiro UFV 2148-57 x H511-1. As inoculações foram feitas pela aplicação de 20 gotas de 5 [micra]l de uredosporos na face inferior de folhas destacadas, no interior de gerbox. Após incubação e limpeza da face inferior da folha inoculada os gerbox foram transferidos para câmara com ambiente controlado. Foram feitas quatro avaliações, iniciadas aos 30 dias após a inoculação, com intervalo semanal. O método permitiu detectar genótipos resistentes e susceptíveis no material analisado, sendo que o controle ‘Catuaí’ apresentou grau de susceptibilidade máxima. O método não apresentou escape e permitiu analisar um grande número de genótipos com uma pequena quantidade de uredosporos. Possui ainda as vantagens de praticidade operacional, mão-de-obra e tempo reduzidos, menor gasto de tecido vegetal e redução da contaminação.

The most striking difficulty to do genetic studies involving the relation of coffee - Hemileia vastatrix is the absolute need of host cell. Therefore, the objective of this work was to evaluate a reliable method to inoculated H vastatrix on coffee to study races and the inheritance resistance of coffee to leaf rust. For that, it was used the race II of H. vastatrix, C. arabica ‘Catuaí’, as the susceptible host, and the coffee segregant population UFV 2148-57 x H511-1. The inoculations were done by applying 20 drops of 5 [micra]l of uredospores on the abaxial surface of detached leaves, maintained in the gearbox. After the incubation, the abaxial surfaces of the inoculated leaves were cleaned and the gearboxes were transferred to a controlled environment chamber. Four evaluations were done, initiated at 30 days after inoculation at weekly intervals. The method permitted to detect resistant and susceptible genotypes in the analyzed material and the control ‘Catuaí’ showed a maximum susceptibility index of disease. The procedure proved to be reliable and very sensitive; it allowed to analyze a labor, it is not time-consuming, requires a small amount of plant tissue and avoids contamination.
